Generating single cell genomics data from primary human samples for translational research is very challenging given that samples must be fresh (to preserve nucleic acid integrity) and large (to accommodate sorting and preparation by FACS). These challenges are uniquely eliminated by combining: 1) newly optimized methods for single nucleus isolation from cryopreserved tissues and 2) the CellRaftA microscale polystyrene growth surface within an array used to spatially segregate cells/colonies while maintaining shared media access, enabling imaging over time and targeted isolation.
